Iron concentrations are elevated in the airways of patients with inflammatory lung diseases such as cystic fibrosis and ARDS. The presence of iron in the airway catalyzes the production of highly reactive oxygen species (ROS) that cause cellular injury and trigger oxidant-mediated intracellular signaling. To limit oxidative stress, iron acquisition and its distribution must be tightly regulated. Airway epithelial cells normally take up iron and sequester it within ferritin to protect the lung’s extracellular milieu; however, many of these mechanisms remain unclear. The goals of our work are to more clearly define mechanisms of cellular iron transport in airway epithelial cells and to determine the role of disrupted iron homeostasis in the pathogenesis of inflammatory lung disease.
