Diagnostic consultation in blood cancers (leukemias), lymphomas and other blood diseases, flow cytometric immunophenotyping, cell subset analysis in HIV infection, cytogenetic and molecular diagnosis of congenital disorders and cancers, diagnostic consultations in diseases of the liver and biliary systems

My research interests are in the area of lymphomas and leukemias.
1. Biology of variant sub-populations in acute leukemia: Multiparameter flow cytometry coupled with cell sorting provide a very powerful tool to analyze small subpopulations in acute leukemia cells. Such subpopulations are defined based on aberrant expression of certain surface antigens. The biological behavior of these subpopulations is largely unknown. Acute lymphoblastic leukemia, the commonest cancer of childhood, often shows variable levels of expression of non-lymphoid antigens on some of the leukemia cells. Conversely, acute myeloid leukemias show variable expression of lymphoid antigens on a proportion of leukemic cells. Whether these differences represent true subclones is not clear. Multiparameter flow cytometry will be used to identify a subset of these cases where the aberrant expression occurs on a distinct sub-population. These cells will be isolated by cell sorting and compared to the dominant population as regards growth rate, cytogenetic abnormalities, growth factor requirement if any, in-vitro response to cytotoxic drugs and expression of selected oncogenes. These cases will be clinically followed up to examine the fate of these subpopulations following therapy and when it occurs, in relapse.
2. Lymphoid lesions of the ocular adnexa: We are analyzing a large collection of lymphomas occurring in and around the eye. The marginal zone lymphomas of the MALT type will be studied extensively to determine the morphological, immunophenotypical, and cytogenetic properties that can predict behavior. In particular, the cytogenetic abnormalities associated with poor outcome in gastric MALT lymphomas will be studied in archival tissues using fluorescent in-situ hybridization and PCR.
3. Biology of non-myeloablative allogeneic stem cell transplantation: Increasing numbers of hematological, lymphoid, and solid tumors are being treated with this approach using reduced intensity conditioning regimens. I am interested in correlating morphological examination of the bone marrow following transplantation, the level of donor engraftment as judged by molecular methods, and patient outcome.
